Twenty four clones were evaluated to sugar cane mosaic virus strain D (SCMV-D) by mechanical inoculation in the glasshouse in Morocco. A field trial established in Malaga, Spain towards the same end, but with twelve varieties, had two blocks, one of which was mechanically inoculated, while in the other one virus transmission was by vectors, presumably aphids. Assessment was by visual observations and virus presence in symptomless plants was assessed by indirect ELISA. In the glasshouse test on the basis of type of symptoms and per centage infected plants, 9 varieties were rated as highly resistant , as they were symptomless, 3 moderately resistant, 6 susceptible and 3 very highly susceptible. The virus was detected by indirect ELISA in four of the nine varieties without symptoms revealing that they were latent carriers of the disease. Under field conditions, mechanical inoculation resulted in much higher disease uptake, up to 75.1 per cent plants infected in variety L7120, than by natural transmission through which maximum infection was 19.4 per cent in the same variety The same trend in varietal reaction was however observed by the two methods. Final survey showed that 4 varieties were highly resistant, 1 resistant, 1 moderately resistant, 2 susceptible, 1 highly susceptible and 2 very highly susceptible. It concluded that varieties can be screened with confidence in the glasshouse and that infection in symptomless plants must be diagnosed by ELISA to identify latent carriers. Such varieties and those which are susceptible to very highly susceptible to SCMV-D, should not be released. However, if they have been so prior to the tests like CP65357, close monitoring is required to avoid them from acting as damaging virus reservoirs in the field. | |
