The exploitation of polymorphisms by an increasing number of molecular marker techniques is starting to have an impact on plant genome research and breeding. Restriction fragment length polymorphisms, polymerase chain reaction-based techniques and mini-satellites are used to determine inter- and intra-specific genetic diversity and to construct molecular maps of crops using especiaily designed mapping populations. Resistance genes and other agronomically important loci are tagged with tightly linked DNA markers, by using highly saturated linkage maps, and genes isolated by mega-base DNA technology and c1oning in bacterial artificial chromosome (BAC) libraries. Molecular markers have considerable impact on breeding of agronomically important crops, because they provide, together with genetic engineering techniques, access to unavailable genetic resources for crop improvement. DNA molecular approaches are being used to identify markers linked to quantitative trait loci (QTLs) associated with yellow spot disease resistance. The goal is to establish a saturated linkage map for specific populations produced by bi-parental crosses. Field experiments were carried out over a period of three years and the disease severity (per cent) for ail individuals was recorded. When one or more markers for an interesting trait are identified and cloned, they could be used to identify resistant individuals derived from a cross, solely by the markers' distribution pattern in the genome of the offspring. This will help plant breeders to identify desirable breeding material in a very short time. | |
