Three specific races of Xanthomonas campestris pv. vasculorum, the gumming disease pathogen, have been identified, mainly from varietal reaction and cultural characteristics of isolates on a special agar medium containing triphenyl tetrazolium chloride. Since differentiation based on cultural charactersitics is time-consuming, more sophisticated techniques were investigated with the aim of finding a rapid and reliable method. Of these, the only one that allowed race differentiation was Restriction Fragment Length Polymorphism (RFLP) using endonucleases and probes; this advanceds biotechnological technique is described and results obtained with it are reported. Since the three races revealed were very closely related, new probes of much lower molecular weight were developed and are being evaluated. It is hoped that it will be possible to monitor the three races precisely as well as new ones before they reach the epidemic stage and cause a varietal breakdown. | |
