The effect of explant source and genotype on growth of sugar cnae in vitro was examined. Explants from three sources, axillary bud and shoot apex were culture. Severe bacterial contamination occurred in axillary buds resulting in necrosis and death of the explants. Growth responses were better with apical buds than with axillary buds in two of the three varieties cultured. In these varieties, B 6504 and M 554/79, 8.3 per cent and 12.5 per cent of axillary buds respectively formed shoots. On the other hand, 26.3 per cent and 36.4 per cent of apical buds developed into shoots. No significant difference in shooting response was detected between axillary buds and shoot apices. Four varieties were cultured to study the influence of genotype on shoot development and significant differences were detected. Best response was obtained with variety M 261/78, with more than 66 per cent of explants developing into shoots. Varieties M 2350/79 and M 529/76 gave almost similar responses with about 37 per cent of the buds forming shoots. M 52/78 responded poorly. | |
