A fragment from the genomic DNA of a strain od Xanthomonas albilineans, previously serotyped as African, has been cloned following subtractive hybridization with a Mascarene strain. This fragment was used as a probe for restriction fragment length polymorphism (FRLP) analysis of different strains of pathogen. Two bands of approximately 3 kb and 6 kb were observed with all the strains tested while a band of 1.3 kb was obtained only with some of the African strains of Mauritius. This probe, F1, was partly sequenced and primers were designed for amplification by polymerase chain reaction (PCR). PCR was carried out on genomic DNA of X. albilineans using the primers derived from the sequence of probe F1. At low primer concentration, a single band of 330 bp was amplified from teh strains of Mauritius serotyped as African. Twenty out of thirty one isolates from other countries produced similar results with primers. No amplification was obtained with the Mascarene serotype of mauritius. With a higher primer concentration a slightly larger band of 370 bp was obtained with some of the latter strains. The primer set F1 A and F1 B can be used to differentiate between strains of X.albilineans belonging to the African and Mascarene serotypes. | |
