Yellow leaf disease of sugarcane caused bySugarcane yellow leaf virus (SCYLV) is widely present in Mauritius. Transmission is primarily through the use of infected planting material, though the involvement of aphids as vectors is well documented. Hot water treatment is ineffective in the control of the virus. Thus tissue-culture techniques are being applied for elimination of the virus. In order to ensure that plantlets being produced are free fom the virus, it is important to have reliable diagnostic tests with high level of sensitivity. A real-time Taqman reverse transcription PCR has been adopted for screening of tissue culture plantlets. SCYLV is known to be genetically diverse and three genotypes were initially described worldwide (BRA-PER, CUB and REU), but recent reports indicate that the variety collection in Mauritius using RT-PCR and a survey of commercial fields has been initiated to assess the diversity. At the same time, real-time RT-PCR assays are being tested for each genotype. Initial results for the specific detection of the CUB genotype are presentedin this poster. Future work will focus on the development of molecular tools for the detection of all genotypes of SCYLV and the possibility of multiplexing, assessing the reaction of commercial cultivars to the virus and inheritance studies. Eventually, it would be possible to develop cultivars resistant to SCYLV and this is a better option for long-term sustainable management of the pathogen. | |
