| Three races of Xanthomonas campestris pv vasculorum (Xcv) the causal agent of gumming disease have been identified in Mauritius using conventional methods based on evolution of the disease, varietal reaction, and cultural properties of the pathogen. More efficient and powerful techniques using monoclonal antibodies, restriction fragment length polymorphisms (RFLPs) and random amplified polymorphic DNA (RAPD) have been adopted for advanced studies on the relationship of isolates of the gumming bacterium present in sugar cane and on alternative hosts such as palm, broom bamboo and maize. Specific hybridomas that allow distinction of Xcv from other bacterial pathogens of sugar cane, including leaf scald X. albilineans, were obtained, as well as race 1 specific monoclonals. Race 1 could be distinguished from races 2 and 3 using DNA probes. The potential of using RAPD which is based on the polymerase chain reaction was also demonstrated. Although no consistent banding pattern on agarose gel could be correlated to the races, hybridisation of amplified products to blotted DNA on filters revealed that race 1 is different from races 2 and 3. These technologies have provided evidence that race 1 is serlogically and genetically different from the two other races. Studies are being persued to separate races 2 and 3 which appear to be more closely related. The ultimate aim of these investigations is to develop tools for epidemiological studies and rapid disease diagnosis. | |
