Abstract (AB)Bacterial wilt of potato caused by Burkholderia solanacearum E. F. Smith (=Pseudoonas solanacearum E F Smith) is a widespread pathogen in Mauritius. Characterization of isolates has shown that the bacterium belongs to race 1 biovar III. As this race is both seed and soil-borne, it is important to detect the bacterium in both environments to ensure that seed produced are clean and that plantation is effected in fields that do not harbour the pathogen. Four techniques were evaluated to that effect: plating on semi-selective media (TZC medium compared to two modified Granada and Sequeira (GS) media), microtitre plate ELISA, nitrocellulose membrane ELISA (NCM-ELISA) and indirect immunofluorescence (IF). Detection in seed was carried out from a piece of tissue obtained from the stolon end of the tuber macerated in sterile distilled water. The diffusates were then used in the various tests. All four methods could detect B. solanacearum in both environments. However, plating was the most sensitive technique. Isolation from soil was most efficient on the modified GS medium supplemented with crystal violet (50 ppm) and polymyxin B sulfate (100 ppm), tyrothricin (20 ppm), chloromycetin (5 ppm) and cycloheximide (5 ppm). Kelman's TZC medium was least effective although it could be used for isolation from tubers. The sensitivity of the plating method was confirmed using a pure culture and was found to be 10²cells/ml on the three media. The level of detection by plate ELISA, NCM-ELISA and IF was 10 6, 10 6 and 10 4 cells/ml respectively.
