Abstract (AB)The sugarcane yellow leaf virus (SCYLV) that is associated with one form of sugarcane yellow leaf syndrome can be detected by both serological and molecular methods. However, many of these tests are tedious when carried out on a large scale, especially with regard to sampling and sample preparation. Most of the available methods use the leaf as the starting material, for which grinding is necessary. An alternative starting material, sugarcane juice, is proposed for stalks having internodes. A simple punching tool is used to collect samples of juice from the stalk internodes and these samples are ready for use in ELISA after a short centrifugation. It was found that there was generally a higher virus titre at the top of stalks compared to the base. SCYLV was present in all the stalks of infected stools. Attempts to cure infected seedcane of the virus by thermotherapy were unsuccessful.
