Abstract (AB)Molecular markers are important tools in various genetic and genome analyses. They cover a wide range of genetic markers revealed by different molecular techniques. The first application of molecular markers started about 30 yrs ago with the use of isozymes. Then restriction fragment length polymorphisms (RFLPs), which started in the late 1980s, allowed a refinement of the taxonomic analysis as well as the construction of detailed genetic linkage maps in many species. The polymerase chain reaction (PCR) using arbirarily selected primers became established in 1990 as a rapid, efficient, and economical method of generating markers that are useful in genetic mapping and genome fingerprinting. In the mid-1990s the development of another PCR-based technique, the amplified fragment length polymorphisms (AFLPs), has gained much success since it allows a progeny-specific detailed map to be made within a very short time compared to other approaches. Microsatellites are also becoming availablefor major crops such as sugar cane. The DNA fragment polymorphism generated by DNA molecular marker techniques can be used: for variety identification and determination of parentage, to score natural populations to study population structure and genetics, as markers to study phylogenetic relationships, and as markers to construct genetic maps from segregating population. The availability of a wide array of markers for plant breeders will allow marker-assisted genetics and selection to be envisioned in the near future.
