Abstract (AB)The maize mosaic virus (MMV) in Mauritius and elsewhere is a rhabdovirus transmitted with low efficiency by the delphacid plant hopper Peregrinus maidis Ashmead. On the basis of the striping pattern induced in infected plants, three strains were identified and designated as MMV-Fine, MMV-Coarse and MMV-Broken. Studies on the histopathology, virus-vector-host relationships, host range, and epidemiology of the three disease types are described. The use of resistant varieties proved to be the best method for control of the disease. Highly purified virus preparations obtained with a procedure involving high speed centrifugation, column chromatography and sucrose density gradient centrifugation enabled partial characterization of the virus particles associated with the three strains of disease. Their sedimentation coefficient did not differ significantly. Highly specific low-titred antisera were prepared for the three isolates, which were found to be serologically identical in straight immunodiffusion tests and in intragel absorption tests. With the ELISA technique, virus antigen could be detected in crude sap up to a dilution of 1:20 000. No difference of size, morphology and ultrastructure of particles associated with the three MMV strains was evident in negatively-stained preparations and in ultra thin sections in situ. The work revealed that the disease described as maize line virus in Kenya is the same as MMV-Broken.
