| A coryneform bacterium has regularly been observed in centrifuged diffusates from tissues of canes showing typical mature node symptoms of ratoon stunting disease in Mauritius. It was not present in healthy symptomless canes originating from cuttings treated in hot water at 50°C for two hours. On the basis of its peculiar morphology, the bacterium is probably the same which has been found associated with the disease elsewhere. It has been obtained from mature nodes and internodes, from apical tissues of mature stalks and young tillers as well as from leaves. This indicates systemic infection rather than a passive localized saprophytic infection through wounds or leaf scars. The bacterium has not been found in stalks affected by chlorotic streak, gumming or leaf scald diseases. Bacteria in diffusates from the latter two diseases were morphologically quite distinct from the one associated with ratoon stunting disease. Examination was done by phase contrast microscopy on smears negatively stained with nigrosine, as well as by electron microscopy. Results obtained with light microscopy on smears negatively stained with nigrosine, as well as by electron microscopy. Results obtained with light microscopy indicate that this method could be useful for routine diagnosis of RSD if the association of the specific bacterium is confirmed. | |
