Abstract (AB)Weatherley's micro-osmometer was used in this study to obtain estimates of diffusion pressure deficits of maize leaves. Determinations of soil suctions with the same apparatus were compared with those obtained with tensiometers and gypsum blocks.The micro-osmometer was calibrated over sodium chloride solutions where readings recorded after a temperature equilibrium time of twenty minutes were found to be constant and linearly related to the concentrations of sodium chloride used. The readings obtained when leaf strips were used suggested that the water in the leaf could be held in a two phase system as was suggested by Strugger , Levitt , Carr and Gaff , and Weatherley. The measurements gave two possible values of leaf DPD . The first one was obtained by extrapolating the rate of evaporation of the drop of water in the osmometer to zero time. the second estimate was taken when a steady rate of evaporation had been reached after two hours . It is suggested that the extrapolated value ( D1) is a measure of the diffusion pressure deficit of the cell wall and the equilibrium value (D) that of the vacuole although there is evidence to the contrary which is discussed. Experiments comparing D1 and D of leaves of plants subjected to various matric soil suctions are described and the possibility of the existence of a polar resistance between the vacuole and the cell wall is discussed.
