Abstract (AB)Different hot water treatments and sterilizing agents were evaluated with the aim of reducing latent bacterial infection in sugar cane buds used in tissue culture work. The Australian cold soak hot water treatment (cold soak in running water for 48 hr followed by hot water treatment at 50°C for 3 h on the following day) of setts prior to bud excision considerably reduced the level of contamination in cuttings derived from a commercial field. This treatment did not have the same effect on cuttings issued from tissue culture canes when compared to untreated controls. Partial elimination of bacterial contamination was achieved with surfece sterilizing agents like sodium hypochlorite at 2.7 per cent (pH 6.0) for 20 min and 1 per cent mercuric chloride for 2 min. Eighty-three per cent of clean cultures were obtained by these tow methods, but both were found to be phytotoxic to the buds. While using germinated and non-germinated buds for in vitro culture, it was found that a higher level of contamination was present in germinated cuttings. Bacterial contaminants were found to be endophytic and were located in both the cutting and the bud. Identification of these organisms was possible using Fatty Acid Profiles and bacteria belonging mainly to the Enterobacter and Bacillus groupd were identified. Only one plant pathogen, Pseudomonas gladioli, were obtained.
