Abstract (AB)The potential of monoclonal antibodies to provide a rapid diagnostic test for the differentiation of the three races of Xanthomonas campestris pv vasculorum (XCV) was investigated. Balb/c mice were immunized by single or repeated intraperitoneal injections of washed, formaldehyde-fixed bacterial suspensions of the three races of the bacterium. A lipopolysaccharide (LPS) extract from bacterial cells was also used as immunogen. A titre of 1:3200 or above was obtained when mice were given a single injection whereas, with the repeated immunizations, titres as high as 1:102400 were obtained in indirect ELISA tests using horseradish peroxidase as conjugate. Fusions of spleen cells with non-secretor myeloma cells X63/Ag8/653 or NSO were performed three days after a final boost with the appropriate antigen. The fusogen consisted of PEG 1500 and addition of 10 per cent dimethyl sulphoxide resulted in a better fusion rate. Hybrid production varied from 10 per cent to 93 per cent. Specific hybrids to XCV were obtained. However, no clones specific to any of the three races were obtained although some showed a slight specificity to each of the three races. Investigations aimed at obtaining specific hybrids secreting antibodies to the three races of XCV are being pursued.
