Abstract (AB)Gumming disease, caused by the bacterium Xanthomonas campestris pv vasculorum (Xcv) is an important disease of sugarcane and in Mauritius three races of the bacterium have been identified. Hybridoma technology was used for the production of monoclonal antibodies for the characterization of isolates. Balb/c mice were immunized by intraperitoneal injections with various antigen preparations namely live cells, heat fixed cells, formaldehyde fixed cells and lipopolysaccharide (LPS) extracts . Specific hybridomas to Xcv were obtained when spleen cells from mice immunized with heat fixed cells or LPS extracts were fused. One fusion produced hybridomas specific to race 1 isolates when compared to the other two races in indirect ELISA and dot blot test. The sensitivity of the specific ascites was found to be 1 x 106 cells/mL in indirect ELISA.
