Abstract (AB)A genomic library was produced following subtractive hybridisation between DNA from Xanthomonas albilineans (serovar I) and Xanthomonas campestris pv. vasculorum. After screening the cloned fragments, probe 83 was selected for its specificity, since it hybridised only to DNA of X. albilineans and not to other DNA tested. With Bam H1 diests of X. albilineans DNA, probe 83 produced three hybridisation profiles, one of which comprised all the Mauritius isolates. It could also differentiate among isolates of X. albilineans classified into two DNA groups by Alvarez et al. This probe was sequenced. Two primers, S83A and S83B were designed and used for PCR, and they produced a single amplification product of ca. 300 p from genomic DNA of 38 isolates of X albilineans tested, originating from sixteen countries or sttes. These primers amplified a band of the same size from DNA extracts of sugar cane leaves infected with the pathogen. Highly reproducible results were obtained both with the PCR using S83A and S83B and the hybridisation with probe 83, suggesting that they can be useful tools for the identification of X. albilineans and for studying genetic variation within ghe species. The availability of such specific primers will help in the control measures for leaf scald disease.
